The long-term vision of our group is to decipher the molecular mechanisms of G protein-coupled receptor (GPCR) allostery for rational drug discovery. There is immense therapeutic potential in the ability to tune receptor signaling using partial agonists, biased agonists or allosteric modulators – and it remains virtually untapped. Our current understanding of biased agonism derives primarily from cell-based functional assays that are easily complicated by receptor expression levels, off-target effects and cell type. Integrating biophysical studies of purified protein with this cellular information will permit a complete molecular understanding. Our work aims to employ a combined biophysical, biochemical, cell-based, and computational approach to characterizing the role of both Monod-Wyman-Changeux (MWC) and dynamically-driven (DD) allosteric mechanisms in the context of GPCR and arrestin activation.

experience are highly desired, with at least an interest in state-of-the-art spectroscopy is required.
